Insert Design Tool for the pSilencer™ Vectors
Step 1. Enter your a) siRNA Target Sequence OR b) the actual siRNA sequence:

a) siRNA Target Sequence (must be 21 bases):
5'- -3'


b) Sense siRNA Strand:
5'- -3'

Antisense siRNA Strand:
5'- -3'

Step 2. Enter your Loop Sequence (Default is shown):

Step 3. Choose the Vector for which you would like to design inserts:

pSilencer 2.0, 2.1, 3.0, 3.1, and 5.1 vectors
pSilencer 1.0-U6
pSilencer adeno 1.0-CMV
pSilencer 4.1-CMV vectors
Other Pol III promoter vector (generates insert sequences with termination sequence, but without restriction site overhangs)

Overview
This tool will generate hairpin siRNA-encoding DNA oligonucleotide insert sequences from an input siRNA target sequence, for cloning into a pSilencer siRNA Expression Vector. The program will add the loop sequence and overhangs for cloning as outlined in the vector Instruction Manual.

Instructions
1) Enter the target sequence to which your siRNA is directed. 2) Use the default loop sequence as shown, or alter the loop sequence as desired. 3) Choose the vector for which you are designing an insert so that oligonucleotide templates are designed with the appropriate restriction enzyme overhangs. Choose "Other" to generate insert sequences without any restriction site overhangs. 4) Click "Submit" to generate the oligonucleotide insert sequences.
