Phage isolation

Greetings virologists,

As a bacteriologist, I am seeking some assistance in the isolation of
a rather interesting phage. We have a phage which appears to be non
virulent and is exuded from the bacterium into the culture medium.
There is no evidence that the DNA genome is intergrated into the host
genome. We are looking for methods that will allow us to increase
phage yield in order for us to have enough particles for DNA
isolation.

Any and all ideas on the best methods for increasing the yield of the
particles AND in isolating the DNA are welcome. Please reply by email
to either myself or Yolanda Petersen at, yolanda at molbiol.uct.ac.za

Many thanks.
