primer extension

Hi, all, 

I am working with an entomopoxvirus. I have done 5' primer extension to make 
certain the start point of a late viral gene. But no good result. 

Question 1 is that if some specific mRNA structure of poxvirus gene can effect 
primer extension result?

Question 2 which key steps should be paid more attention in primer 
extension experiment?
